Section titled “1. -Leu Sc Media Plates”- 250 mL of 4% agar solution (10g) and autoclave for 20 minutes on liquid cycle.
- 250 mL of 2X -Leu SC media: 0.81 g -leu dropout mix, 3.35 g YNB, 10 g glucose.
- Heat with stirring until very warm.
- Filter sterilize.
- Mix -leu SC media with 4% agar solution.
- Wait until cool enough and pour onto plates.
2. YES Plates

- Autoclave for 15 minutes on liquid cycle.
3. SC-Ade Plates

| Yeast Nitrogen Base w/o Amino Acid | 3.35 g | |
| agar | 7.5g | |
| H2O | to 500 mL |
- Autoclave 15-20 minutes on liquid cycle.
- Pour plates- be careful about contamination as there is no antibiotic to prevent growth on the plates.
